138 Single-cell RNA-seq of dorsal root ganglion reveals neuroinflammatory process in atopic dermatitis

Atopic dermatitis is a chronic inflammatory skin disease, the pathology of which characterized by barrier dysfunction, microbiome shift, immune dysregulation initiated severe itching sensation. Because its pathophysiological and clinical significance, research on understanding mechanism itch finding new therapeutic targets it has been highlighted recently. To dissect cellular/molecular changes associated with itch, we obtained total six mouse dorsal root ganglion tissues; three from atopic dermatitis-induced NC/Nga mice another control mice, performed single-cell RNA sequencing. Among non-neuronal cells, proportions neutrophil macrophage increased in dermatitis, implies presence neuroinflammation. Non-peptidergic neurons, have nociceptors transmitting sensation, occupy larger proportion among neurons compared to control. As first transcriptome analysis DRG, our study provides insight into how sensation manifests itself single cell level dermatitis.
